Preparation of edible antimicrobial packaging

This bachelor’s thesis is focused on preparation and characterization of eatable packaging. Eatable packaging consists of two components, antimicrobial component and base of packaging. The task of eatable packaging is to protect product against antimicrobial attack and to extend durability of the product. As antimicrobial components were used water and oil extracts of several herbs (mint, nettle, lavender, cinnamon, cloves, vanilla, ginger and rhinoceros). Oil extracts were also encapsulated into liposomes for better application. Firstly, concentrations of polyphenoles and antioxidants of extracts were determined. Oil extract of clove contained the highest concentration of both determined substances. Encapsulation efficiency, stability and size of liposomes were tested too. All prepared liposomes were stable and relatively of the same size. Also high encapsulation efficiency was observed. Next, antimicrobial activity of prepared extracts and liposome particles against yeast Candida glabrata and two bacteria strains Escherichia coli and Staphycococcus epidermidis was tested and the high antimicrobial activity reported mainly all tested liposomes. Then alginate, chitosan and carboxymethylcellulose and their combinations respectively were used as bases of packaging. Created film had to meet several parameters, especially suitable sensory properties such as pleasant taste and smell, as well as invisibility, gloss, affordability and suitable manipulability. Combination of 2% carboxymethylcellulose and 1% alginate in a ration of 2:1 with liposomes of mint as antimicrobial component achieved the best results.

The diploma thesis deals with the issue of herbal tea, which is registered as medicinal product. The consumer market research has shown that these kinds of tea are put on the market by Megafyt Pharma s.r.o. and Leros s.r.o. An overview of all single-species medicinal tea and tea mixtures from both producers was created. In the practical part, the content of essential oil in the medicinal tea leaves of chamomile and sage from the producer Megafyt Pharma s.r.o. and the content of tannins in the tea leaves of Agrimonia eupatoria from the same producer were determined. The samples of individual drugs of different ranks were used for the determination and the evaluation was performed according to the Český lékopis 2017. The results were statistically evaluated. The obtained results showed that the essential oil content of chamomile corresponds to the pharmaceutical quality, but for the sage sample the essential oil content does not correspond to the given standard. In the case of Agrimonia eupatoria, the content of tannins was lower than determined by the Český lékopis 2017, only one sample of the given rank showed a higher content. Furthermore, the frequency of occurrence of individual species of herbs contained in the registered tea mixtures was evaluated.

This thesis deals with the analysis of herbs traditionally used for the absinth production: Pimpinella anisum, Foeniculum vulgare and Artemisia absinthium. The suitable GC-MS methods were developed and validated and they were used for the determination of the volatile substances content in essential oils and dried herbs which are available at the Czech market. Another aim of this study was to determine the content of t anethole and -thujone during the distillation of absinth in both laboratory and industrial scale. Finally, distillation residues were tested on Spodoptera litoralis larvae for potential pesticidal effects at VÚRV.
Biological activities of Lavandula angustifolia essential oil

The thesis called "Biological activities of Lavandula angustifolia essential oil" is interested in biological activities of Lavandula angustifolia essential oil, specifically antifungal, antioxidant, anti-inflammatory, cytotoxicity, nematicidal and repellency activities. The aim of work was to find out, if the oil can be used in these indications and become natural substitution to the chemical compound toxic to the environment. Antifungal activity was tested against the most common human patogenes, low toxicity proves that essential oil can be used on human cells without causing any harm. Antioxidant activity was tested on NO reaction, nematicidal activity was tried on proctection of crop where nematodes cause huge economic loss. Repellency activity can protect both decorative and agricultural plants. Quick determination of compounds contained in caraway (Carum carvi L.) by a method usable in agricultural practice

Caraway is a very important agricultural commodity whose quality is determined by parameters such as dry matter, essential oil content and composition, especially the ratio of its two major components carvone and limonene. Appropriate method for their analysis is given in the Cesky lekopis (2017), however, this method is rather time-consuming, costly and demands large quantity of sample. The use of a NIR spectrometer could be a viable alternative, it is much faster and cheaper, as can be clearly seen from the comparison of both methods in this paper. In the time aspect, it's saving from many hours to a few minutes. Newly presented method could potentially be more accessible to agricultural companies who need quick quality verification of their product before taking it to the market from the viewpoint of final product quality not just the quantity, which, in most cases, is nowadays a current state of practice.

Microbicidal properties and chemical composition of essential oils

The microbicidal properties of 6 essential oils (EOs; Lavandula angustifolia, Cymbopogon nardus, Citrus aurantifolia, Juniperus communis, Myrtus communis and Cinnamomum zeylanicum ) for 17 microorganisms were determined using the vapour-agar contact method. The most effective EO (i.e. Lavandula angustifolia ) whose volatile components provided the sufficient microbicidal properties was chosen for detailed study of chemical composition.
